She was born into a Brahmin Hindu family in Ernakulam, Kerala.

She started learning Carnatic music at the age of 5 under Dr. Malini Hariharan and Tamarakkadu Govindan Namboodiri.
While she was studying in school and college, she won various singing competitions. In college, she won the State Winner Award in Carnatic Music for the MG University Youth Festival.
She also learned to play Sarangi under T. H. Subramaniam.
She won first prize in classical singing and violin competitions in the Kerala State Level Youth Festival for five consecutive years (i.e. 2001–2005).
She completed her graduation and post-graduation with distinction.
Roopa Revathi has won the musical talent hunt reality show Super Star Global. The show was aired on Amrita TV.
On 1 May 2010, after her marriage to the Indian singer Dr. Kuldeep Muralidhar Pai, she moved to Chennai.
Some of her music albums are Devagayakan (2009), Paarinu Meete (2011), Kamakshi Mahimai (2014), Kamakshi Kavacham (2014), and Meere Paadu Nee (2017).
She has sung songs in a few Tamil films such as Thaamarai Pookkaley (2011), Kadhal Raagam (2011), A-Vandhu B-Male Modhi (2012), and Dharmam Vazha (2012).
She has lent her voice to many Malayalam films like Omanichumma (2012), Akkare Ikkare (2015), Ottathooval (2015), and Olangal Moodum (2017).
In 2013, she joined the music band of Chitra Chechi.
In 2017, she started uploading her cover songs videos on her various social media accounts.
On her self-titled YouTube Channel, she uploads her instrumental cover versions of popular songs.
