He joined the Bharatiya Janata Party (BJP) in the early 1990s during the Ram Janmabhoomi movement.
In 1993, Jishnu Dev Varma was appointed as the national general secretary of the BJP.

In the 2018 Tripura Legislative Assembly election, he contested and won the Charilam seat by over 25,000 votes, receiving 89% of the 29,753 valid votes cast in the by-poll. He defeated his opponent Arjun Debbarma of the Indian National Congress.
Jishnu Dev Varma was also designated as the Deputy Chief Minister of Tripura during this term.
He handled the portfolios of Power, Rural Development (including Panchayat), Finance, Planning and Coordination, Science, Technology, and Environment.
Apart from being a politician, Jishnu Dev Varma is also an author who has published many books including Indigenous Wisdom- Man and Nature (2022), Maharaja Bir Bikram College: A Dream College (2022), Con (Fusion) Of Language and Origin (2023), and The Children of Water Goddess (2024).

He often contributes his articles and poems to various newspapers in Tripura.
On 22 September 2022, Jishnu Dev Varma was appointed as Chairman of the North East Regional Power Committee (NERPC).
In the 2023 Tripura Legislative Assembly election, he was the richest candidate from Tripura. He lost the election to Subodh Deb Barma of the Tipra Motha Party.
In 2023, he was the richest sitting MLA of Bhartiya Janta Party with assets worth Rs. 11 crores.
On 27 July 2024, President Droupadi Murmu appointed Jishnu Dev Varma as the Governor of Telangana. He began his term as Governor on 31 July 2024.
